The joke is that the Sole Survivor thinks she thinks she’s human at first. You’re reaching really hard for some sort of transphobia here. And her voice isn’t masculine. It’s androgynous at best.
And if you’re going to pull the “designation” line, the actual line is: “New Designation: K-L-E-0 ‘Kleo’. Fully independent small business owner.“ Meaning her programming has changed. Because, y’know, she’s a robot.
